Abstract
    To assess whether regulation of steroidogenesis during artificial maturation resembles that in the wild, we compared mRNA levels of three ovarian steroidogenic enzymes (P450 17 alpha -hydroxylase C17-20 lyase (P450c17), 17 beta -hydroxysteroid dehydrogenase Type I, and aromatase) in artificially maturing Japanese eels (Anguilla japonica; JE) and naturally maturing New Zealand longfinned eels (A. dieffenbachii; LFE). Using northern blot analysis, transcripts for all three enzymes were detected in mid-vitellogenic (MV) ovaries of artificially maturing JE. However, transcripts were almost undetectable in previtellogenic (PV) ovaries of JE, and in both pre and mid-vitellogenic ovaries of wild LFE. Using newly established, high-sensitivity competitive PCR, relative P450c17 transcript levels in artificially maturing JE were about five times higher than in the remaining groups. These findings suggest that steroidogenic enzyme genes are overexpressed during artificial maturation of JE, possibly due to the large quantities of hormone injected.
