Abstract
Hemocyanins from 4 spp of decapod crustaceans: Panulirus regius (PR), Neptunus validus (NV), Macropipus puber (MP) and Maia squinado (MS) have been purified by preparative zone electrophoresis in Pevikon. Immunoelectrophoresis of the isolated hemocyanins performed with a specific antihemolymph immune serum reveals one precipitin line. Hemocyanin dissociation by SDS in the presence or absence of a reductive agent gives subunits with a mol wt (MW) close to 80,000. The MW estimation performed by SDS polyacrylamide gel electrophoresis gives the following mean values: 78,000 for NV hemocyanin (NVH), 80,000 for MPH and 82,000 for PRH and MSH. Fingerprinting of these hemocyanins shows the existence of an important percentage of common peptides between the 4 studied hemocyanins. The number of common peptides increased in relationship with the systematic position of the animals and can reach 80% when the Neptunus validus and Macropipus puber hemocyanins are compared. These results confirm the existence of common antigenic determinants described in a previous paper.
