The possibility to develop a routine batch culturing method for the production of cysts has been examined at laboratory scale. The influence of abiotic and biotic parameters on the mode of reproduction has been studied and the following facts have been noted: - cyclic low oxygen tensions induce haemoglobin synthesis with subsequent shift of the mode of reproduction from ovoviviparity to oviparity; - addition of ferric-EDTA stimulates haemoglobin synthesis;- a correlation exists between the color of the animals and the type of reproduction; - neither the food source nor the water salinity per se control the mode of reproduction. However, cysts produced at low salinity appear to have a low hatchability. The effect on the hatchability of various processing techniques applied to the cysts produced has been tested. From these experiments it appears that: - the cysts must be activated (dehydrated) to allow hatching at a subsequent hydration; - the method of dehydration influences to a large extent the viability of the cysts. Of the various methods tested out so far. drying at 40°C or immersion in a saturated brine solution gave the best results. A modified version of the classic air-water-lift operated raceway culturing-system is proposed as a standard method for the controlled production of cysts.
